چکیده

26 Pathological synchronization of neuronal firing is considered to be an inherent property of 27 epileptic seizures. However, it remains unclear if the synchrony increases for the high frequency 28 activity as well as for the local field potentials. We present spatio-temporal analysis of 29 synchronization during epileptiform activity using wide band (up to 2000 Hz) spectral analysis 30 of multi-electrode array recordings at up to 60 locations throughout the mouse hippocampus in 31 vitro. Our study revealed a prominent structure of LFP profiles during epileptiform discharges, 32 triggered by elevated extracellular potassium, with characteristic distribution of current sinks and 33 sources with respect to anatomical structure. The cross-coherence of high frequency activity 34 (500-2000 Hz) across channels was reduced during epileptic bursts compared to baseline 35 activity, and showed the opposite trend for lower frequencies. Furthermore, the magnitude of 36 cross-coherence during epileptiform activity was dependent on distance; electrodes closer to the 37 epileptic foci showed increased cross-coherence and electrodes further away showed reduced 38 cross-coherence for high frequency activity. These experimental observations were recreated and 39 supported in a computational model. Our study suggests that different intrinsic and synaptic 40 processes can mediate paroxysmal synchronization at low, medium and high frequencies. 41

Dynamics of high-frequency synchronization during seizures.

Pathological synchronization of neuronal firing is considered to be an inherent property of epileptic seizures. However, it remains unclear whether the synchrony increases for the high-frequency multiunit activity as well as for the local field potentials (LFPs). We present spatio-temporal analysis of synchronization during epileptiform activity using wide-band (up to 2,000 Hz) spectral analysi...

HIGH-DOSE ORAL PYRIDOXINE FOR TREATMENT OF PEDIATRIC RECURRENT INTRACTABLE SEIZURES

Intractable epilepsy is a common clinical problem in pediatrics and approximately 13% of children with epilepsy experience intractable seizures. To determine the efficacy of pyridoxine in treating seizures, 30 infants and children with recurrent seizures were enrolled in the present study. All of them were treated with high-dose oral pyridoxine (40 mg/kg/day), as an adjunct to antiepileptic...

Development of hypersynchrony in the cortical network during chemoconvulsant-induced epileptic seizures in vivo.

The prevailing view of epileptic seizures is that they are caused by increased hypersynchronous activity in the cortical network. However, this view is based mostly on electroencephalography (EEG) recordings that do not directly monitor neuronal synchronization of action potential firing. In this study, we used multielectrode single-unit recordings from the hippocampus to investigate firing of ...

Hippocampal EEG and behavior: changes in amplitude and frequency of RSA (theta rhythm) associated with spontaneous and learned movement patterns in rats and cats.

Slow electrical activity was recorded from the dorsal hippocampus in rats during running in a motor-driven wheel, swimming, conditioned avoidance (running, jumping), lever pressing for food and sleep, and in cats during walking in a treadmill, eating and lapping milk. Large-amplitude clear rhythmical slow activity (RSA) was recorded from the hippocampus proper and smaller amplitude RSA and fast...

A semi-automated algorithm for studying neuronal oscillatory patterns: a wavelet-based time frequency and coherence analysis.

In many experimental designs, animal observation is associated with local field potential (LFP) recordings in order to find correlations between behavior dynamics and neuronal activity. In such cases, relevant behaviors can occur at different times during free-running recordings and should be put together by the time of analysis. Here, we developed a MATLAB semi-automated toolbox to quantitativ...
